IL-15 maintains T cell survival via inhibition of caspase-3-mediated T cell apoptosis

Active caspase-3 is an effector cysteine protease crucial for apoptosis. T lymphocyte survival impinges on the ability of the cell to keep the levels of active caspase-3 in check. This seminar will focus on how cytokines interleukin (IL)-2 and IL-15 differentially regulate the survival of T cells. Previous studies have shown that despite IL-2 and IL-15 sharing γc and IL-2Rβ (CD122) receptor subunits and being capable of stimulating proliferation and activation of T cells, these two cytokines provide contrasting contributions to T cell life and death. The mechanism for this is not well understood. Data presented will show that the distinct effects on T cell survival by these two cytokines are due to differential regulation of caspase-3 activity independent of caspase-8 and -9 activation.
